免疫组织化学（石蜡）

A. 溶液与试剂

1. 二甲苯。
2. 乙醇，无水变性，组织学分级（100% 和 95%）。
3. 去离子水 (dH₂O)。
4. Hematoxylin（可选）。
5. 洗涤缓冲液：
   1. 1X Tris Buffered Saline with Tween^® 20 (TBST): 要制备 1L 1X TBST，添加 100 ml 含 Tween^® 的 2010 X Tris 盐缓冲液 (#9997) 至 900 ml dH ₂0，然后进行搅拌。
6. 抗体稀释剂： SignalStain^® Antibody Diluent (#8112)
7. 1X EDTA Unmasking Solution: To prepare 250 mL of 1X EDTA unmasking solution, dilute 25 ml of SignalStain^® EDTA Unmasking Solution (10X) (#14747) with 225 mL of dH2O.
8. 3% 过氧化氢： 要制备，添加 10 ml 30% H₂O₂ 到 90 ml dH₂O 中。
9. 封闭液：TBST/5% Normal Goat Serum 或 1X Animal-Free Blocking Solution。
   1. TBST/5% Normal Goat Serum：向 5 ml 1X TBST 中添加 250 µl Normal Goat Serum (#5425)。
   2. 1X Animal-Free Blocking Solution：在 4 mL 的 dH₂O 中，加入 1 ml 的 Animal-Free Blocking Solution (5X) (#15019)。
10. 检测系统：SignalStain^® Boost IHC Detection Reagent (HRP, Rabbit #8114)。
11. 基质：SignalStain^® DAB Substrate Kit (#8059)。
12. Hematoxylin（苏木精）‭： Hematoxylin(#14166)。
13. 封片剂： SignalStain^® Mounting Medium (#84583)。

B. 脱蜡/复水

注：务必使切片在制作过程中始终保持湿润状态。

1. 脱蜡/水化合步骤：
   1. 将切片放入二甲苯的洗液中孵育 3 次，每次 5 分钟。
   2. 将切片放入 100 乙醇中孵育切片 2 次，每次 10 分钟。
   3. 将切片放入 95 乙醇中孵育切片 2 次，每次 10 分钟。
2. 用 dH₂O 清洗切片两次，每次 5 分钟。

C. 抗原修复

注：本流程描述了使用 Biocare Medical Decloaking Chamber 所建议的条件。设备专用设置和操作说明应用于其它加压蒸煮器。

1. 将切片放入 250 ml 室温存放的 EDTA 修复液中，切片放在一个能放 24 个切片的切片夹上。
2. 将 500 ml dH₂O 倒入加压蒸煮器中。
3. 将切片夹放入加压蒸煮器，直至接触到隔热板。再将一个能放 24 个切片的切片夹放入有 250 ml 水和空白切片的加压蒸煮器中，这样也许有好处。
4. 密封蒸煮器室，并继续进行修复。以下为 Biocare Medical Decloaking Chamber 的设置。
   1. SP1 125°C 30 秒
   2. SP2 90°C 10 秒
5. 小心给设备开排放口，随后取下盖子并将切片放在实验台上冷却 10 分钟。
6. 用 dH₂O 冲洗切片。

D. 染色

1. 用 dH₂O 清洗切片三次，每次 5 分钟。
2. 切片放入 3% 过氧化氢水溶液中，孵育 10 分钟。
3. 用 dH₂O 清洗切片两次，每次 5 分钟。
4. 用洗涤缓冲液清洗切片 5 分钟。
5. 在每个切片上滴加 100–400 µl 首选封闭液，在室温下封闭 1 小时。
6. 清除封闭液，并向每块切片添加 100-400 µl 在 SignalStain^® Antibody Diluent #8112 中稀释过的一抗。4°C 孵育过夜。
7. 使 SignalStain^®Boost Detection Reagent (HRP, Rabbit8114#) 平衡至室温。
8. 清除抗体溶液，并用洗涤缓冲液清洗切片 3 次，每次 5 分钟。
9. 按需在切片上滴 1-3 滴 SignalStain^® Boost Detection Reagent (HRP, Rabbit 8114#)。在加湿箱中室温孵育 30 分钟。
10. 用洗涤缓冲液清洗切片 3 次，每次 5 分钟。
11. 加入 1 滴 (30 µl) SignalStain^® DAB Chromogen Concentrate 至 1 ml 的 SignalStain^® DAB Diluent 中，使用前充分混合。
12. 在每张切片上应用 100–400 µl SignalStain^® DAB 并密切观察，通常 1–10 分钟即可得到合适的染色强度。
13. 将切片浸入 dH₂O 中。
14. 如果需要，使用 hematoxylin (14166#) 复染切片。
15. 用 dH₂O 清洗切片两次，每次 5 分钟。
16. 使切片脱水：
    1. 在 95% 乙醇中孵育切片 2 次，每次 10 秒。
    2. 在 100% 乙醇中重复孵育 2 次，每次 10 秒。
    3. 在二甲苯中重复孵育 2 次，每次 10 秒。
17. 使用盖玻片和封片剂 (84583#) 封片。
