Protocol

Specificity / Sensitivity

TdTomato (E3G5L) Rabbit mAb detects transfected and transgenic levels of TdTomato-tagged proteins (either N-terminally tagged or C-terminally tagged). Please note that the TdTomato tags add approximately 54.2 kDa to the molecular weight of the fusion protein.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of TdTomato protein.

Background

Epitope tags are useful for the labeling and detection of proteins using immunoblotting, immunoprecipitation, and immunostaining techniques. Because of their small size, they are unlikely to affect the tagged protein’s biochemical properties.

TdTomato is a fluorescent, photostable orange dye that was isolated from and further engineered from the Discosoma sea anemone (1). Reported to be six times brighter than EGFP, TdTomato is comprised of a tandem dimer of monomers engineered from DsRed; the dimers are held together by intramolecular bonds. For this reason, TdTomato is considered a monomer. Its peak fluorescent excitation is at 554 nm and emission is at 581 nm (1).
