    Source / Purification

    Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val361 of human sortilin protein. Antibodies are purified by peptide affinity chromatography.

    Background

    Sortilin is a multifunctional member of the vacuolar protein sortin-10 (VPS10) receptor family containing type-1 transmembrane proteins that have pleiotropic function in neuronal protein trafficking and viability (1). Sortilin is implicated to have inhibitory effects on tau aggregation in Alzheimer's disease transgenic murine models expressing human mutated tau P301S (2), and also supports to regulate intracellular processing of amyloid precursor proteins (APP) (3,4) and β-secretase, which cleaves APP to produce neurotoxic Aβ peptides (5). Sortilin also plays a role in frontotemporal lobar degeneration (FTLD) by binding and regulating extracellular concentrations of the secreted glycoprotein progranulin (6). Sortilin has been linked to cholesterol circulating concentration and lifetime risk of developing significant atherosclerotic disease. Higher concentrations of sortilin have been found in patients with coronary atherosclerosis (7). Sortilin functions in the immune system within microglia, pro-inflammatory cytokine regulation, phagosome fusion, and pathogen clearance (8).
