    Phospho-PKC Substrate Motif [(R/K)XpSX(R/K)] MultiMab® Rabbit mAb mix detects endogenous levels of many cellular proteins only when phosphorylated at Ser residues surrounded by Arg or Lys at the -2 and +2 positions. The antibody may also cross-react with S*X(R/K) motif-containing proteins. The antibody does not cross-react with nonphosphorylated Ser residues, phospho-Thr in the same motif, or phospho-Ser in other motifs.

    MultiMab® rabbit monoclonal mix antibodies are prepared by combining individual rabbit monoclonal clones in optimized ratios for the approved applications. Each antibody in the mix is carefully selected based on motif recognition and performance in multiple assays. Each mix is engineered to yield the broadest possible coverage of the modification being studied while ensuring a high degree of specificity for the modification or motif.

    Background

    Although protein kinase C (PKC) family members are involved in a number of signal transduction processes including secretion, gene expression, proliferation, and muscle contraction, many PKC substrates continue to be unidentified (1,2). Isozymes of PKC are subdivided into conventional PKCs (cPKC), novel PKCs (nPKC), and atypical PKCs (aPKC). PKCα, βI, βII, and γ isoforms belong to the cPKC group (1). When activated, cPKC isozymes phosphorylate substrates containing Ser or Thr, with Arg or Lys at the -3, -2, and +2 positions, and a hydrophobic amino acid at position +1 (1-3).
