    Phospho-GRASP55/GORASP2 (Thr264) Antibody recognizes endogenous levels of GRASP55/GORASP2 protein only when phosphorylated at Thr264. This antibody may detect a nonspecific band around 45 kDa.

    Source / Purification

    Polyclonal antibodies are produced by immunizing animals with a synthetic phosphopeptide corresponding to residues surrounding Thr264 of human GRASP55/GORASP2 protein. Antibodies are purified by peptide affinity chromatography.

    Background

    In unconventional protein secretion (UPS), cytoplasmic cargo proteins that do not have a signal peptide or a transmembrane domain are transported to the plasma membrane or extracellular matrix mainly to respond to cellular stresses (1,2). The Golgi reassembly-stacking protein 2 (GRASP55, also known as GORASP2) is essential to type III and type IV UPS (2,3). Studies show that mTORC1 phosphorylates GRASP55/GORASP2 and keeps it at the Golgi apparatus under non-stressed conditions. When cells are stressed, mTORC1 is inhibited, leading to dephosphorylation of GRASP55/GORASP2. Dephosphorylated GRASP55/GORASP2 relocates to autophagosomes and multivesicular bodies to promote UPS. In addition, Thr264 on GRASP55/GORASP2 was shown to seemingly play a key role in mTORC1-mediated phosphorylation of the protein (2).
