    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Leu703 of human PDE4A protein.

    Background

    Phosphodiesterase (PDE) superfamily enzymes catalyze the hydrolysis of 3',5'-cyclic nucleotides into the corresponding nucleotide 5'-monophosphates. The PDE superfamily includes 11 subfamilies (PDE1-PDE11) in mammals (1). These enzymes function as important positive and negative regulators of cellular response, including regulation of insulin secretion, heart function, erectile function, and inflammation (2-5). The PDE4 subfamily consists of PDE4A, PDE4B, PDE4C, and PDE4D isoforms, and are cAMP-specific enzymes expressed in T cells, monocytes, macrophages, dendritic cells, neutrophils, and eosinophils (6). PDE4 expression is induced by epithelial damage, allergen sensitization, and microbial invasion (7). Inhibition of PDE4 subfamily members leads to the elevation of intracellular cAMP and modulates the inflammatory response (8). The isoform, PDE4A, is being investigated as a therapeutic target for various inflammatory conditions such as asthma, inflammatory bowel disease (IBD), and rheumatic arthritis (RA), and as an immune checkpoint inhibitor for multiple suppressive factors (9,10).
