    Protocol

    Specificity / Sensitivity

MRCKα (E2S5I) Rabbit mAb recognizes endogenous levels of total MRCKα protein. Based on the sequence of the peptide antigen, this antibody is not expected to cross-react with MRCKβ or MRCKγ.

    Species Reactivity:

    Human, Mouse, Rat

    Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Gly1728 of human MRCKα protein.

    Background

CDC42BPA/MRCK (Myotonic dystrophy kinase-related CDC42-binding kinase) comprises a family of serine/threonine kinases that function as downstream effectors of the small GTPase, CDC42. Three MRCK family members have been identified: MRCKα, MRCKβ, and MRCKγ. MRCK family kinases have some substrates in common with the Rho-associated kinases, ROCK1 and ROCK2, including Myosin Light Chain 2 (MLC2). Signaling through both ROCK and MRCK kinase families regulates actin dynamics, cell migration, invasion, and metastasis (1,2). Simultaneous inhibition of both ROCK and MRCK kinases prevents migration and invasion of human cancer cell lines (3). Small molecule inhibitors targeting the MRCK family are being investigated as potential treatments for glioblastoma and skin cancer (4,5).
