    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the amino terminus of human MCT4/SLC16A3 protein.

    Background

    MCT4, also known as MOT4 or SLC16A3, is a multi-pass transmembrane monocarboxylate transporter (MCT) protein encoded by the gene SLC16A3 (1,2). MCT4 is reported to be expressed in white (glycolytic) muscle cells (3,4), astrocytes (5), activated macrophages (6), and cancer cells (2). The primary role of MCT4 is lactate exportation, which is adapted to the release of lactate from glycolytic cells (8,9). In the tumor microenvironment, MCT4 mediated lactate secretion promotes M2 macrophage polarization and suppresses T cell function (10). MCT4 inhibition increases the efficacy of immune checkpoint blockade and is a potential target to reverse tumor resistance to immunotherapy (11,12).
