    Background

    Mab-21 domain-containing protein 2 (MB21D2), also known as C3orf59, belongs to a protein family of cGAS/DncV-like nucleotidyltransferase (CD-NTase) enzymes involved in a variety of cellular processes, including cell survival, proliferation, and migration (3). MB21D2 is an intracellular E-cadherin binder that carries a recurrent Q311E mutation (1,4). Both wild-type (WT) and Q311E mutant MB21D2 have been identified as being frequently overexpressed in lung, head, and neck cancers, and may mediate pro-oncogenic activities by promoting epithelial-mesenchymal transition (EMT)/cancer stemness (2-4).
