    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Ala194 of human COL2A1 protein.

    Background

    COL2A1 (α1 type II collagen) is a fibril-forming collagen encoded by the gene COL2A1 (1,2). The protein forms homotrimeric type II collagen and is the main extracellular component in hyaline cartilage extracellular matrix (ECM), nucleus pulposus of intervertebral discus, vitreous humor of the eye, and inner ear structure (3). Type II collagen plays a fundamental role in maintaining cartilage formation, differentiation, and growth (4-6). Mutations of COL2A1 cause impairment of ECM deposition and signaling, which contribute to various forms of tissue dysplasia and pathologies such as Stickler syndrome and chondrosarcoma (3,7,8). At the ECM, COL2A1 interacts with integrins such as integrin β1 and growth factors such as IL1β, IL6, IL8, and others, which activate intracellular signaling pathways, including SMAD1, and MAP kinases p38, JNK, and ERK (9-12).
