    Background

    DNA-(apurinic or apyrimidinic site) endonuclease 2 (APEX2/APE2) is an endodeoxyribonuclease that functions in the DNA base excision repair (BER) pathway to repair DNA lesions induced by oxidative and alkylating agents. APEX2 acts in ATR-dependent DNA damage checkpoint activation in response to oxidative stress (1). The APEX2 gene is synthetic lethal with BRCA1 and BRCA2 genes, which are required for homologous recombination (HR) repair of double-stranded DNA breaks. This is due to the ability of APEX2 to reverse single-stranded DNA breaks with 3’ end blocks. Because BRCA-deficient cancer cells are sensitive to 3’ blocks, APEX2 is a potential therapeutic target for HR-deficient cancers (2,3).
