    UNC-2170 Hydrochloride is supplied as a lyophilized powder. For a 15 mM stock, reconstitute 5 mg of powder in 0.95 mL of DMSO. Working concentrations and length of treatment can vary depending on the desired effect.

    Storage

    Store lyophilized at -20ºC, desiccated. In lyophilized form, the chemical is stable for 24 months. Once in solution, store at -20ºC and use within 3 months to prevent loss of potency. Aliquot to avoid multiple freeze/thaw cycles.

    Background

    UNC-2170 Hydrochloride is a small-molecule inhibitor of p53-binding protein 1 (53BP1), a protein involved in DNA damage response. By screening a set of methyl-lysine binding proteins, UNC-2170 was identified as a fragment-like ligand that preferentially binds 53BP1 (1). 53BP1 is a methyl-lysine binding protein recruited to double-strand DNA breaks, playing a central role in DNA damage repair by promoting non-homologous end joining (NHEJ) repair (2). The 53BP1 protein contains a tandem tudor domain responsible for the recognition and binding of methylated lysine residues on histone proteins. Because UNC-2170 binds the same 53BP1 domain recognized by histones (Kd = 22 ± 2.5 µM), UNC-2170 competes for histone binding and functions as a 53BP1 antagonist in cellular lysates (1). UNC-2170 is only a modestly potent inhibitor of 53BP1 protein function; UNC-2170 analogs and other compounds that bind and inhibit 53BP1 are under development (3).
