Stattic is supplied as a lyophilized powder. For a 40 mM stock, reconstitute 10 mg of powder in 1.18 ml of DMSO. Working concentrations and length of treatment can vary depending on the desired effect.

Storage

Store lyophilized at -20ºC, desiccated. In lyophilized form, the chemical is stable for 24 months. Once in solution, store at -20ºC and use within 3 months to prevent loss of potency. Aliquot to avoid multiple freeze/thaw cycles.

Background

Stattic is a nonpeptidic selective Stat3 inhibitor. This small molecule prevents the binding of tyrosine-phosphorylated peptide motifs to the Stat3 SH2 domain, inhibiting Stat3 activation, dimerization, and nuclear translocation with an IC50 value of 5.1 μM (incubated for 1 hour at 37°C) (1). Stattic blocks IL-6-induced Stat3 activation, reducing cell viability in tumor cells (2). Treatment with 10 μM induces apoptosis in Stat3-dependent cancer cell lines MDA-MB-231 and MDA-MB-435S (1).
